| FRAP

FK506-binding protein 12-rapamycin-associated protein 1 (FRAP1) also referred to as the mechanistic target of rapamycin and mammalian target of rapamycin (mTOR),s a kinase that in humans is encoded by the MTOR gene.mTOR is a member of the phosphatidylinositol 3-kinase-related kinase family of protein kinases.mTOR integrates the input from upstream pathways, including insulin, growth factors (such as IGF-1 and IGF-2), and amino acids.mTOR also senses cellular nutrient, oxygen, and energy levels. The mTOR pathway is a central regulator of mammalian metabolism and physiology, with important roles in the function of tissues including liver, muscle, white and brown adipose tissue, and the brain, and is dysregulated in human diseases, such as diabetes, obesity, depression, and certain cancers.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 20 10 5 2.5 1.25 0.62 0.31 0
OD 2.19 1.61 1.05 0.66 0.43 0.29 0.21 0.07
Corrected OD 2.12 1.54 0.98 0.59 0.36 0.22 0.14 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human FRAP spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Tissue Homogenates(n=8) 84-101 96
Cell Lysates(n=8) 92-105 102

| Linearity

Samples were spiked with high concentrations of Human FRAP and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Tissue Homogenates(n=4) Cell Lysates(n=4)
1 : 2 Range (%) 84-95 89-96
Average (%) 91 93
1 : 4 Range (%) 89-103 87-108
Average (%) 94 98
